Bio-fuels are a fairly new thing here in the UK and information is hard to come by.
If I'm using E5 should I be setting the AFR in General Settings to 14.4 rather than 14.7?

No, the wideband can't taste the difference between alcohol, gasoline or propane for that matter, just the free oxygen%
Internally widebands are all lambda devices, so it will show you stoich for gasoline (as they are ~always setup for that) at lambda 1.0...
... regardless if its E0, E5 or E100.
I worried about this too until that finally registered.
E5 isn't really different enough to worry about anyway, the ms3 ethanol sensor //table mixing setup doesn't alter anything until after E10.
I'm not sure what that TunerStudio stoich setting is for, really. It's kinda senseless. Just leave it at 14.7:1.
Maybe error correction from a funky sensor setup?
There is a custom.ini here somewhere for setting up a proper lambda gauge.
We all should really be using that anyway.
The firmware does.

Addendum to Newtons first law:
zero vehicles on jackstands, square gets a fresh 090 and 1911, cabby gets a blower.
EZ3.6 Vanagon after that.(mounted, needs everything finished) then Creamsicle.
