Please Help with this clutch problem

My mum has an Isuzu ks32 1984 bus. She has just replaced the pressure plate and now she has no clutch. She is wanting know if the bus needs to be going for the pressure to build up to operate the clutch.

This is a Volkswagen forum, there's nobody here with any special knowledge regarding Isuzus so far as I know. That vehicle probably never shipped anywhere beyond Oceania, either, so you're only likely to find folks who are knowledgeable on it in New Zealand, Australia, and nearby countries.
When you find a forum which does discuss Isuzus, you should specify what is meant by "no clutch". Does that mean that it will not disengage to allow shifting with the engine running, or that there is no transmission of power through it? Does the pedal action feel normal when depressed?
I'm also unclear as to what you mean by "the bus needs to be going" ...I presume you mean the engine must be running? I know of no vehicle which relies upon the engine to run for the clutch release mechanism to operate. If the KS32 has a hydraulically operated release bearing mechanism and the line was disconnected in order to remove the transmission and replace the pressure plate, there will be air in the line which needs to be bled, similar to bleeding brakes, before the clutch will disengage.
